چکیده

It has been demonstrated that solid hepatoma lysosomes lack hyaluronidase activity, whereas normal rat liver lyso somes contain a high level of this enzyme. This result was also found with lysosomes from an ascites hepatoma, Walker carcinoma 256, and virally transformed BHK cells. In contrast, 0-yV-acetylglucosaminidase activity was shown to be present in lysosomes from tumor cells, although its level was decreased by about 75%, compared with that of controls. As it is known that hyaluronidase and /3-/V-acetylglucosaminidase activities are increased in the extracellular space of tumors, we suggest that the lack of hyaluronidase and the important decrease of /8-,/V-acetylglucosaminidase in tumor lysosomes are due to a perturbation of the control of enzyme distribution from the Golgi apparatus to the lysosomes and the extracellular space.

Activities of some enzymes concerned with citrate and glucose metabolism in transplanted rat hepatomas.

1. Certain enzymes concerned with citrate and glucose metabolism have been measured in two transplanted rat hepatomas, one induced with ethionine (minimal deviation type) and one induced with dimethylaminoazobenzene. In these hepatomas both citrate-cleavage enzyme and NADP-linked isocitrate dehydrogenase in the soluble fraction of the cell were approximately one-third of the values for normal r...

RNA sulfurtransferase activity in rat liver and chemically induced hepatomas.

RNA sulfurtransferase activity has been detected in rat liver and in hepatomas from rats fed a diet containing 0.06% 3'-methyl-4-dimethylaminoazobenzene for 14 to 18 weeks. The reaction measured was the transfer of sulfur from cysteine to acceptor sites in Escherichia coli B transfer RNA (tRNA). Specific activities of the enzymes in liver and hepatoma supernatant fractions were similar, as were...

Diamine oxidase activity in regenerating rat liver and in 4-dimethylaminoazobenzene-induced and Yoshida AH 130 hepatomas.

Diamine oxidase (EC 1.4.3.6) activity, measured as delta 1-[14C]pyrroline formation from [14C]putrescine, was studied in homogenates of regenerating liver and of 4-dimethylaminoazobenzene-induced by Yoshida AH 130 hepatomas of rat. The addition in the incubation medium of acetaldehyde increased delta 1-pyrroline formation in normal and regenerating liver that contained aldehyde dehydrogenase bu...

Preparation of single-cell suspensions from normal liver, regenerating liver, and Morris hepatomas 9121 and 5123tc.

Suspensions of single cells were prepared from normal adult rat liver either by mincing the tissue and incubating it in various concentrations of sodium tetraphenylborate, ethylenediaminetetraacetate, and 0.1% hyaluronidase plus 0.05% collagenase or by continuous recirculating perfusion of the liver with 0.1% hyaluronidase plus 0.05% collagenase. Only the latter method produced cells in good yi...
